L’équipe Chimie Organique et Interfaces (CORINT) est l’une des composantes de l’Institut des Sciences Chimiques de Rennes (UMR CNRS 6226). Elle regroupe 70 chercheurs, enseignants-chercheurs, ingénieurs, personnels techniques et administratifs de l’ENSCR, de l’Université de Rennes 1 et de l’INSA Rennes. Les principaux objectifs de recherche visent à concevoir et produire des composés organiques à haute valeur ajoutée, de développer de nouvelles voies de synthèses et d’étudier les interactions qui gèrent de nombreuses applications aux frontières de la biologie et de la physique.

Les recherches de l’équipe CORINT s’organisent autour de 5 axes :

  • Les produits naturels et les molécules pour le Vivant (produits marins et issus de lichens, métabolites, sucres, lipides, enzymes).;
  • Méthodologies, outils moléculaires et synthèse (nouvelles réactions multi-composants, réactifs bi-métalliques, chimie du bore, chimie prébiotique et interstellaire) ;
  • Matériaux moléculaires dédiés (assemblages supramoléculaires, formulation, vectorisation mais aussi composants optiques pour la thérapie photodyamique et composants optoélectroniques) ;
  • Structures and interactions (études des interactions dans des milieux complexes ; RMN, ITC, …)
  • Technologies et chimie éco-responsable (mise en œuvre des principes de la chimie verte, biocatalyse, et procédés hybrides)

Parallèlement au nécessaire accroissement des connaissances fondamentales, les retombées visées se trouvent principalement dans les domaines de la santé (anti-cancéreux, antiparasitaires, maladies du système nerveux central).

A l’ENSCR, la quinzaine de permanents est plus particulièrement impliquée dans les thématiques liées à :

  • La conception de molécules pour des fonctions données. Cet axe comporte un volet hétérocomposés oxygénés et azotés, notamment les isonitriles et les oxydes de nitrile. Les plus simples de ces composés présentent un grand intérêt pour une meilleure connaissance du milieu interstellaire, des atmosphères planétaires et des queues de comètes. L’interface avec le domaine de l’exobiologie est actuellement développée. Glycochimie et biocatalyse constitue le second volet de cet axe. Il s’agit de concevoir et de synthétiser des glycoconjugués bioactifs d’intérêt notamment pour des applications dans le domaine de la santé humaine (immunostimulants, antiparasitaires). Les approches chimio-enzymatiques visent à remplacer une étape classiquement catalysée chimiquement par un biocatalyseur conçu à façon ;
  • La conception d’architectures moléculaires pour des fonctions définies. Le premier volet est dédié à La vectorisation de principes actifs et le transfert de gènes. L’équipe est depuis plusieurs années impliquées dans la synthèse et l’étude d’analogues de lipides membranaires d’archaebactéries dans le but de fournir de nouveaux vecteurs de principes actifs, pour des applications en cosmétique et en santé humaine (thérapie anti-cancéreuse notamment). Les formulations produites sont par ailleurs basées sur des polymères biocompatibles visant essentiellement le traitement de cancers hépatiques. Le second volet est relatif à l’imagerie et la radiothérapie métabolique impliquant la vectorisation de radioisotopes utilisés en imagerie fonctionnelle. Le troisième volet concerne des molécules amphiphiles issues de matières premières renouvelables. Ces travaux ont deux objectifs : la valorisation de matières premières renouvelables à travers des applications en cosmétologies et dans l’industrie routière.

Synthèse de composés cinétiquement instables

De nombreux hétérocomposés n’ont jamais été synthétisés à cause de leur instabilité cinétique. Cependant, si leur réactivité peut être contrôlée, nombre d’entre eux pourraient devenir des composés clés en chimie organique grâce à leur polyfonctionnalité. Une partie de nos recherches est dédiée aux boranes, complexés ou non. Néanmoins, la majeure partie de nos études concerne des dérivés oxygénés et azotés. Ainsi, les nitriles, les isonitriles et les oxydes de nitriles représentent le plus grande partie de notre activité. De plus, les plus simples de ces composés présentent un grand intérêt pour une meilleure connaissance du milieu interstellaire, des atmosphères planétaires et des queues de comètes. L’interface avec le domaine de l’exobiologie est actuellement développée. Ici sont présentés quelques résultats récents de notre groupe.

Synthèse d’hétérocomposés liés à l’astrochimie

– Synthèse de cyanopolyynes
Les cyanopolyynes, de formule H-(C≡C)n-CN, sont probablement les composés organiques détectés dans le milieu interstellaires les plus intéressants. Cependant, leur synthèse en laboratoire reste un véritable défi à cause de leur instabilité, même si le premier membre de cette famille, le cyanoacétylène (n=1), a été synthétisé en 1910. Notre groupe a été le premier à synthétiser le membre suivant de cette famille, le cyanobutadiyne (n=2).  La réactivité de ce composé a ainsi pu être étudiée pour la première fois, ainsi que les probables mécanismes réactionnels qui ont pu mené à sa formation dans le milieu interstellaire et éventuellement dans l’atmosphère de Titan.

– Synthèse d’adduits du cyanoacétylène
Le cyanoacétylène, premier membre de la famille des cyanopolyynes, est souvent considéré comme un composé clé de la chimie prébiotique qui s’est développée sur la Terre Primitive. C’est la raison pour laquelle nous avons beaucoup étudié sa réactivité ses dernières années.

– Isonitriles, nitriles, aldéhydes, … des candidats pour le milieu interstellaire
Nous essayons également d’aider les astrophysiciens à détecter de nouveaux composés organiques dans le milieu interstellaire.  Pour ce faire, nous suggérons et synthétisons de nouveaux composés organiques pertinents. Ainsi, les spectres de micro-onde de ces composés sont enregistrés en collaboration avec d’autres groupes dans le but de les identifier dans l’espace in fine.

Synthèse et étude d’hétérocomposés simples

– Synthèse de composés α,β-insaturés
Des nitriles et hétéroacrylonitriles polyinsaturés et isonitriles insaturés ont été synthétisés. Nous essayons actuellement d’utiliser ces composés en synthèse organique pour former, en peu d’étapes, des composés élaborés.

– Synthèse de complexes
Des complexes entre des amines ou des phosphines et des boranes ou des alanes ont été synthétisés et étudiés par spectroscopie. La possibilité pour certains d’entre eux d’être intéressants pour le stockage d’hydrogène a été développée.  D’autres pourraient être des précurseurs de boranes fonctionnalisés libres.

Propriétés physico-chimiques des hétérocomposés

La synthèse de nombreux nouveaux composés nous a conduit à développer des collaborations nationales et internationales avec des spectroscopistes et des théoriciens pour en avoir une meilleure connaissance. En particulier, l’acidité en phase gazeuse de complexes ou de composés originaux a été étudiée.

Dans un autre domaine, nous avons développé un sujet fondé sur la compréhension de la sublimation partielle de mélanges énantio-enrichis. La possibilité d’obtenir des composés enrichis en un énantiomère à partir d’un racémate, ou d’amplifier un excès énantiomérique par sublimation est actuellement étudiée. Cette thématique est en lien avec la volonté de comprendre l’origine de l’homochiralité sur Terre.

Synthèse de tétracyanobutadiènes

Nous nous sommes récemment intéressés à la séquence de réactions de cycloaddition-rétroélectrocyclisation [2+2] (CA-RE) entre des ynamides et le tétracyanoéthylène. En effet, nous avons réalisé une méthodologie montrant qu’il était possible de créer des tétracyanobutadiènes à partir de plusieurs ynamides et du tétracyanoéthylène en les mélangeant simplement dans du dichlorométhane à température ambiante. Cette réaction est très tolérante à de nombreux substituants. Nous étudions actuellement la possibilité d’étendre cette méthodologie à des systèmes plus complexes et d’en étudier les propriétés en optique non-linéaire.

Furanosides

Les hexofuranosides sont présents dans des biomolécules complexes produites par des microorganismes souvent pathogènes, mais sont absentes chez les mammifères. Compte tenu de la résistance accrue aux antibiotiques et d’une réémergence de maladies telles que la tuberculose ou la lèpre, de l’impact de champignons tels que Aspergillus fumigatus chez des patients immunodéprimés ou de trypanosomes ou leishmanies dans les pays en voie de développement, les furanosides et leurs voies de biosynthèse sont devenues des cibles privilégiées pour élaborer des thérapies ou des outils moléculaires originaux.

Dans ce contexte, notre équipe a développé des stratégies de synthèse de disaccharides et d’oligosaccharides contenant une ou plusieurs unités furanosidiques, ainsi que des aryl- et alkyl-furanosides . Ces réalisations sont basées sur :

  • la chimie des thioimidates pour les réactions de glycosylation chimiques ;
  • l’utilisation d’enzymes hydrolytiques et de thioglycoligases.

Nous avons notamment montré que des dérivés d’alkyl-galactofuranosides présentent une activité antimycobactérienne, ainsi qu’une activité très prometteuse contre des leishmanies.

1,3-glucanes

Parmi les très nombreux polysaccharides naturels, les β-(1,3)-glucanes sont reconnus pour leur potentiel immunostimulant, c’est-à-dire qu’ils sont capables de renforcer le système immunitaire des mammifères. Ils trouvent donc toute leur place comme adjuvants bioactifs dans l’accompagnement de traitements anticancéreux.

Notre contribution dans ce domaine consiste à concevoir et synthétiser des oligo-β-(1,3)-glucanes de structures parfaitement définies, naturels ou présentant des modulations structurales fines. Les approches chimiques ont récemment été complétées par l’apport de méthodes enzymatiques pour la préparation d’intermédiaires clés.

Les propriétés immunostimulantes ont ensuite été démontrées dans le cadre de collaborations nationales ou internationales.

Glycoconjugués

Notre équipe est également spécialisée dans le domaine de la synthèse de glycoconjugués et notamment de glycolipides. Nous nous attachons notamment à les préparer de façon écoresponsable en utilisant la biocatalyse par exemple (Glycosidase ou Lipase).

Certains de ces glycolipides permettent d’accéder à des hydrogels via un auto-assemblage dans l’eau sous la forme de fibres.

D’autres glycolipides sont dérivés du tréhalose et constituent à ce titre des mimes de glycolipides généralement présents chez les organismes pathogènes tels que mycobactérie.

Valorisation de bioressources

On peut citer par exemple la valorisation d’huile végétale en matériaux polymères. Là encore les procédés utilisés sont simples et les plus respectueux de l’environnement. Les materiaux obtenus ont des Tg relativement basse (proche de 0°C) et pourraient trouver des utilités dans l’industrie des plastiques notamment.
